What is StemSource® Stem Cell Bank?

Your healthy stem cells are stored frozen for use in the treatment of diseases in the future.

Stem cells stored frozenStemSource® Stem Cell Bank (cryopreservation system for adipose tissue-derived stem cells) is an advanced medical treatment service in which stem cells collected from your fat are stored frozen in a nitrogen tank at −150°C and will be used for the treatment of diseases, rejuvenation, and breast reconstruction in the future. All cells, including stem cells, age over time, and thus it is important to store stem cells when you are young and healthy.
Clinical studies have been conducted on stem cell-based therapies for intractable diseases and other disorders worldwide, and the efficacy has been demonstrated. Cryopreservation of stem cells collected while you are healthy serves as a kind of insurance; those cells will be used for therapy when you fall ill in the future. Stem cells are stored frozen at −150°C without mycoplasma contamination.Under strict control, those cells are safely stored for a long period of time.

Potential of regenerative medicine with stem cellsRegenerative medicine is a medical treatment intended to regenerate cells, tissues, or organs and recover body functions lost by accidents or disease. There are several types of regenerative medicine, and transplantation of stem cells is one of them. In recent years, stem cells have been extensively studied along with the development of culture technology, molecular cytology, tissue engineering, and genetic engineering. Stem cell-based regenerative medicine is now attracting attention as a national product. Stem cells have been used for the treatment of a variety of intractable diseases, and clinical studies are now ongoing.

Stem cells have the ability to transform into specific cells and renew themselves in an undifferentiated state for a long time. In other words, stem cells are master cells that grow into tissues or organs. The use of your own cells to regenerate tissues or organs and restore function will overcome immunological rejection and ethical problems. Clinical studies have been conducted on stem cell-based therapies for a variety of diseases, and the efficacy has been demonstrated. Advances in stem cell therapy are remarkable, and progress over the past few years suggests that the therapy will surely develop further.

adipose tissue-derived regenerative cells, ADRCStemSource® Stem Cell Bank at our center uses adipose tissue-derived stem cells (adipose tissue-derived regenerative cells, ADRC) harvested from your fat.
Research institutions in Japan and the United States are now investigating embryonic stem (ES) cells collected from human embryos (fertilized eggs) and induced pluripotent stem (iPS) cells prepared by transferring multiple genes into human somatic cells such as skin cells or the amnion. The former uses fertilized eggs and raises ethical problems, and the latter uses a cancer-related gene (*1) and has a problem of an increased incidence of cancer. In addition, study results are reported every day. Because of a limited amount of collectable stem cells and a small number of stem cells, culturing is necessary.
In contrast, the stem cell bank in our center has advantages; the bank overcomes ethical problems by using your adipose tissue-derived stem cells (autologous tissue) and eliminates the need for cultures by using the Celution® 900/MB System.

*1: In May 2009, researchers at Harvard University in the United States successfully prepared iPS cells without the use of genes or viruses with oncogenic potential. However, their procedures require eight weeks, double the period required by conventional procedures, and the efficiency is one-tenth that of standard methods. Their research is now ongoing.

Mycoplasma is a eubacterium, and contamination of cells with the organism is called mycoplasma contaminationDuring the storage of cells, care should be taken to prevent mycoplasma contamination. Mycoplasma is a eubacterium, and contamination of cells with the organism is called mycoplasma contamination. Mycoplasma contamination is sometimes in the news. There is a risk of contamination at universities and hospitals where cells are stored frozen at −80°C to −90°C. The FDA in the United States strictly stipulates in the good tissue practice (GTP) that cells should be stored frozen at −150°C or below to prevent mycoplasma contamination. In the StemSource® Stem Cell Bank at our center, cells are stored in a nitrogen tank at −150°C in accordance with the GTP to ensure safety. We perform all processes from fat collection (liposuction) to cryopreservation in accordance with the Cytori Therapeutics’ dedicated system and safety control manuals (GTP and GMP). All kits used for stem cell extraction are disposable.

There are several cell sources for stem cells including the bone marrow and cord blood. However, the yield of stem cells is much higher in adipose tissue than in any other cell source. This indicates that stem cells are abundant in adipose tissue. Another advantage is that a substantial number of fresh stem cells may be used soon after collection from adipose tissue without culturing. Cord blood and the amnion have the problem of limited donors. However, fat can be collected from anyone, and weight loss is an additional advantage.

Celution SystemCollection of many effective adipose tissue-derived stem cells requires liposuction without damage to adipocytes. Destruction of adipocytes decreases the number of effective stem cells. Seishin Regenerative Medicine has one of the largest number of liposuction cases in Japan, and physicians have one of the best techniques in the industry, as evidenced by study results. A study showed that adipose tissue collected at our center contained a larger number of effective stem cells than that collected at other clinics.

Celution® 900/MB System for safe, automated extraction of stem cells

In the StemSource® Stem Cell Bank at Seishin Regenerative Medicine, the Celution® 900/MB System is used to extract stem cells from your fat. This system was developed by Cytori Therapeutics, which was founded by Dr. Mark Hedrick who discovered adipose tissue-derived stem cells. The system automatically separates, cleans, concentrates, and extracts regenerative cells (ADRC), including effective, reliable adipose tissue-derived stem cells, in approximately one hour, which are ready for storage.
